When designing a dsDNA duplex, it is important to ensure that the sense and antisense oligos are 100% complementary. However, wobble positions and partially non-complementary stretches can be used to create overhangs. It is the responsibility of the customer to specify the sequence of both strands. Unless otherwise specified, the duplex will be produced with hydroxyl groups at both ends. Hylabs can also add modifications, such as phosphate groups, upon request.
The resulting dsDNA oligos are un-cloned, linear DNA fragments that can be used for a variety of downstream applications, such as cloning and crystallization.
