Macroarrays
For high-throughput library screening, gene expression and gene discovery applications, bacterial colonies and DNA can be arrayed at high density on nylon membranes in a custom defined pattern. These membranes can be hybridized to reduce the number of putative positive clones for a secondary screen.
Each colony is present on the membrane in duplicate and can easily be traced back to the well location of your microtiter plate. To facilitate spot identification following the hybridization experiment guide spots are included in each sub-array on the membrane.
After the hybridization screening the membranes can be analyzed by array analysis software or overlays showing the location of your signal.
Up to 55,000 individual colonies (i.e. 27,500 bacterial clones in duplicates) can be spotted onto a 22 x 22 cm positively charged nylon membrane. PCR products can be spotted at even higher density.
Multiple copies of the membranes can be made and delivered ready for radioactive or non-radioactive screening procedures and suitable for stripping and re-probing.
Microarrays
Hy Laboratories Ltd. designs and provides 60 to 70-mers oligonucleotides for your selected genes. These oligonucleotides and control sequences (or cDNA products alternatively) will be spotted in duplicates onto slides with modified glass surfaces or nylon membranes. All oligonucleotides are subject to quality control by mass spectrometry.
- The Total RNA (from all samples) provided by the customer or extracted by our facility is checked prior to the labeling reaction and than labeled with two fluorescent dyes (Cy-3 & Cy-5) or with DIG-11-dUTP when using nylon membranes.
- The molecular biology service lab performs the hybridization reaction.
- The molecular biology service lab performs the scanning and image processing of all microarrays.
Catalog Starter Arrays
Genemed human arrays consists of oligonucleotides representing genes that were selected for non-redundancy from various databases (e.g. EBI, NCBI, Ensemb, GoldenPath). Each individual oligo has been designed using proprietary design algorithms that ensure absolute gene specificity. Powerful bioinformatics delivers one probe per gene.
