DNA synthesis - antisense oligos
Antisense technology has opened a new approach to the development of drug therapeutics. antisense drugs are designed to inhibit the production of disease-causing proteins and offer the potential to be highly selective in their action and less toxic than traditional drugs.antisense oligonucleotides, either as synthetic DNA or RNA, are oligonucleotides which have been modified in various ways. The purpose of this modification is to prevent oligonucleotide degradation within cells, and to increase affinity to the specific target mRNA.
- Phosphorothioates (PTOs): increased cell uptake, nuclease resistance, elicitation of RNAse H activity
- 2´-O-Methyl RNA: resistance to a wide variety of ribo- and deoxyribonucleases, formation of more stable hybrids with complementary RNA strands than equivalent DNA and RNA sequences
- C-5 propyne derivatives: enhancement of duplex stability
The different modifications can also be used in combination with each other to produce chimeric sequences for improved nuclease resistance and to achieve a more potent antisense activity. Performing double purification by HPLC and gel filtration.
Antisense oligonucleotidesPhosphorothioates (PTOs)
Our oligos used for antisense-studies in vivo are fully deprotected, HPLC purified and desalted via cartridge purification! They are free of cytotoxic by-products and ready-to use for cell culture experiments. Due to their modified sugar and/or backbone chemistries antisense oligos are about 100 times more resistant against intracellular nucleases than "normal" DNA oligos so that specific transcription and/or translation inhibition can be very effective. As "Standard"-antisense Oligonucleotides (also called the "first generation oligonucleotide drugs") we offer Phosphorothioates (PTOs) which are the most commonly used today.Chimeric
